Justification for the choice of excipients in the creation of tablets based on pasternax of seeding thick extract

Abstract

Introduction. In modern conditions, cardiovascular diseases occupy one of the leading positions among non‐ communicable diseases worldwide, which in turn leads to disability and mortality. The most common nosological forms that require special attention and immediate action by health professionals are coronary heart disease (with signs of angina and myocardial infarction), atherosclerosis, stroke, heart failure, cardioneurosis, hypertension, peripheral artery disease. A special role in this regard in the treatment of the above pathologies belongs to herbal medicines, the relevance of the development and implementation in production and medical practice of which is beyond doubt.

The purpose: Justification of the choice of excipients for their introduction into the tablets of cardioprotective, anti‐inflammatory and antispasmodic action, developed on the basis of the plant substance of parsnip sowing grass extract dense for the treatment and prevention of diseases of the cardiovascular system.

Materials and methods. Physico‐chemical, pharmaco‐technological and statistical methods of analysis were used in the work. As the active pharmaceutical ingredient, parsnip seed herb extract was selected and the excipients were approved for medical use. Tablets were obtained by wet granulation. Studies of the effect of excipients on such indicators as the homogeneity of the mass of the tablets, their abrasion and resistance to crushing, as well as the disintegration of the tablets were performed in accordance with the requirements of HFC, 2nd edition. The dependence of the resistance of tablets to moisture on the nature of the excipients in their composition was also studied.

Results. The results of the tests indicate the inexpediency of the preliminary conversion of the thick extract into a loose state when mixed with the test media, due to the high values of moisture absorption capacity of the obtained systems. Therefore, in the next test step, the thick extract was directly introduced into the tablet mass, followed by granulation. Based on the conducted pharmaco‐technological studies, we found that the optimal residual moisture content in the mass for tableting should be within (2.5 ± 0.5)%. The optimal value of compression pressure within 110 MPa was determined, which ensures disintegration of tablets within 5–6 min. and the required abrasion at the level of 0.2‐0.3%.

Conclusions. Based on the conducted physicochemical and pharmaco‐technological studies and the obtained results, the optimal composition of excipients was selected for a new tablet drug developed on the basis of the plant substance of parsnip sowing grass extract by the method of wet granulation, namely: microcrystalline cellulose, potato starch XL 10, Aerosil.
